Eldest son of Sanada Masayuki and first lord of the Matsushiro domain. At Sekigahara he sided with the Tokugawa, apart from his father and brother, and in 1622 he moved from Ueda to Matsushiro.
At the Battle of Sekigahara in 1600 he joined the Tokugawa side, parting ways with his father Masayuki and brother Nobushige (Yukimura). He served 11 years as lord of Numata, 23 as lord of Ueda and 35 as lord of Matsushiro, and died in retirement in 1658 at 93.
